Version 5.0.14239 has the Demon Hunter guide update.

If you are asking about when you will be able to play the Demon Hunter in the game you will need to check the Blizzard forums for that answer. I would guess probably after the normal Tuesday maintenance.

I beleive it is Wednesay now.
